Fungal Spore Collection

Fungal spores and hyphal fragments are ubiquitous components of the atmosphere and may occur in high concentrations unless the bottom is roofed with snow or ice. Fungi reproduce by spores, which are produced by either sexual or asexual methods, and the majority of fungal spores are adapted for airborne dispersal.F. oxysporum was maintained on solid PDA medium and after 7 days and the subsequent spore suspension was separated through two layers of sterile cheddar material to dispose of any mycelial pieces and along these lines the conidial suspension was centrifuged at 4,500 rpm, 4ºC for 5 min.The spores were washed threefold with sterile water to evacuate the spore adhesive lastly resuspended in sterile water at a centralization of ×106 spores/ml. The spores were developed for 6 h in PDA stock at 28ºC at 100 rpm.
