Abstract

In vitro regeneration potential of patchouli shoot tip and leaf explants

An effective, reproducible and in vitro rapid regeneration protocol for Patchouli shoot tip and leaf explantswas studied formassmultiplication of plants. The shoot tip cultured on MS (Murashige and SkoogÂ’s) medium regenerated directly in to shoots and the medium supplemented with 2.0 mg l-1 BAP and 0.1mg l-1 NAAexhibited higher percentage of proliferation and registered maximum shoot weight (15.90 g). Callus was established from the leaf explants on the same medium supplemented with varying concentration of 2, 4-D and BAP. 2.0 mg l-1 2, 4-D and 0.1 mg l-1 BAP produced maximumcallusweighing 55.30 g. Regeneration fromthe callus was observed on the same hormonal concentration as for direct regeneration. The regenerated shoots were readily rooted on half strength MS mediumand IAA has had greater influence on number and length of roots. Plantlets readily adapted to greenhouse conditions with 98% of success.
